Men eftersom denna rytm \u00e4r olika f\u00f6r varje person och beror p\u00e5 ett antal olika faktorer \u00e4r det sv\u00e5rt att anpassa l\u00e4kemedelsintaget till patientens individuella kroppsklocka. Forskare vid Charit\u00e9 &#8211; Universit\u00e4tsmedizin Berlin har nu utvecklat en metod f\u00f6r att fastst\u00e4lla den optimala tidpunkten f\u00f6r <a href=\"https:\/\/www.chronobiology.com\/how-chronobiology-is-affecting-cancer-treatment\/\" target=\"_blank\" rel=\"noopener\">cancerbehandling<\/a> baserat p\u00e5 specifika br\u00f6stcancercellinjer. De beskriver sitt tillv\u00e4gag\u00e5ngss\u00e4tt i tidskriften <a href=\"https:\/\/www.nature.com\/articles\/s41467-024-51611-3\" target=\"_blank\" rel=\"noopener\"><em>Nature Communications<\/em><\/a><em>.<\/em> M\u00e4nniskans inre klocka best\u00e4mmer rytmen f\u00f6r m\u00e5nga olika kroppsfunktioner och \u00e4mnesoms\u00e4ttningsprocesser, t.ex. s\u00f6mn och matsm\u00e4ltning. Men det \u00e4r inte bara organen som \u00e4r mer eller mindre aktiva beroende p\u00e5 vilken tid p\u00e5 dygnet det \u00e4r.<\/p>\n<p>\u00c4ven de enskilda cellerna f\u00f6ljer en cykel som best\u00e4ms av den m\u00e4nskliga kroppsklockan, s\u00e5 att de reagerar olika p\u00e5 yttre p\u00e5verkan vid olika tidpunkter p\u00e5 dygnet. Detta \u00e4r av stor betydelse f\u00f6r cellgifter vid behandling av cancer. Tidigare studier har visat att <a href=\"https:\/\/www.chronobiology.com\/chronobiology-and-chemotherapy-drug-timing-improves-treatment-effectiveness\/\" target=\"_blank\" rel=\"noopener\">cellgiftsbehandling<\/a> \u00e4r som mest effektiv n\u00e4r tum\u00f6rcellerna delar sig. Denna uppt\u00e4ckt har dock knappast anv\u00e4nts i klinisk behandling hittills. Ett tv\u00e4rvetenskapligt team p\u00e5 Charit\u00e9 under ledning av Dr. Adri\u00e1n Enrique Granada fr\u00e5n Charit\u00e9 Comprehensive Cancer Center (CCCC) satte sig f\u00f6r att t\u00e4ppa till denna lucka.<br \/>\nTeamet f\u00f6rs\u00f6kte hitta den optimala tidpunkten f\u00f6r att administrera l\u00e4kemedel baserat p\u00e5 tum\u00f6rernas individuella dygnsrytm.<\/p>\n<h2>Exempel p\u00e5 trippelnegativ br\u00f6stcancer<\/h2>\n<p><a href=\"https:\/\/www.chronobiology.com\/wp-content\/uploads\/2024\/08\/can.jpg\"><img decoding=\"async\" class=\"wp-image-63575 size-medium alignright\" title=\"kan\" src=\"https:\/\/www.chronobiology.com\/wp-content\/uploads\/2024\/08\/can-300x180.jpg\" alt=\"kan\" width=\"300\" height=\"180\" srcset=\"https:\/\/dev.chronobiology.com\/wp-content\/uploads\/2024\/08\/can-300x180.jpg 300w, https:\/\/dev.chronobiology.com\/wp-content\/uploads\/2024\/08\/can-768x461.jpg 768w, https:\/\/dev.chronobiology.com\/wp-content\/uploads\/2024\/08\/can.jpg 800w\" sizes=\"(max-width: 300px) 100vw, 300px\" \/><\/a>&#8221;Vi odlade celler fr\u00e5n patienter med trippelnegativ <a href=\"https:\/\/www.chronobiology.com\/being-a-night-owl-increases-breast-cancer-risk\/\" target=\"_blank\" rel=\"noopener\">br\u00f6stcancer<\/a> f\u00f6r att se hur de reagerar p\u00e5 de l\u00e4kemedel som administreras vid olika tidpunkter p\u00e5 dygnet&#8221;, f\u00f6rklarar Carolin Ector, forskningsassistent i Granadas team. Trippelnegativ br\u00f6stcancer \u00e4r en mycket aggressiv form av br\u00f6stcancer f\u00f6r vilken det finns f\u00e5 effektiva behandlingar. &#8221;Med hj\u00e4lp av live imaging, en metod f\u00f6r kontinuerlig \u00f6vervakning av levande celler, och komplexa dataanalystekniker kunde forskarna i detalj observera och utv\u00e4rdera dessa cancercellers dygnsrytm, tillv\u00e4xtcykler och l\u00e4kemedelsrespons. P\u00e5 s\u00e5 s\u00e4tt kunde forskarna identifiera specifika tider p\u00e5 dygnet d\u00e5 cancercellerna reagerar b\u00e4st p\u00e5 l\u00e4kemedelsbehandlingar.<\/p>\n<p>Till exempel fann man att det kemoterapeutiska medlet 5-fluorouracil (5-FU) \u00e4r mest effektivt mot en viss cancercellinje mellan klockan \u00e5tta och tio p\u00e5 morgonen. Som studien ocks\u00e5 visar \u00e4r det vissa cellul\u00e4ra och genetiska faktorer som \u00e4r avg\u00f6rande. Forskarna kunde till och med fastst\u00e4lla vilka gener som \u00e4r avg\u00f6rande f\u00f6r dygnsrytmeeffekten av vissa l\u00e4kemedel. &#8221;Vi kallar dem f\u00f6r &#8217;core clock-generna&#8217;. Enligt Granada har de ett betydande inflytande p\u00e5 hur v\u00e4l cancerceller svarar p\u00e5 behandlingar som ges vid olika tidpunkter p\u00e5 dygnet.<\/p>\n<h2>Profiler visar hur olika typer av cancerceller reagerar p\u00e5 l\u00e4kemedel<\/h2>\n<p>Denna metod kan anv\u00e4ndas f\u00f6r att skapa detaljerade profiler som visar hur olika typer av cancerceller reagerar p\u00e5 olika <a href=\"https:\/\/www.chronobiology.com\/being-a-night-owl-increases-breast-cancer-risk\/\" target=\"_blank\" rel=\"noopener\">l\u00e4kemedel<\/a> vid olika tidpunkter. Enligt forskarna kan detta bidra till att fastst\u00e4lla de mest effektiva l\u00e4kemedelskombinationerna. Sammantaget tyder dessa resultat p\u00e5 att personliga behandlingsplaner baserade p\u00e5 individuella dygnsrytmer avsev\u00e4rt skulle kunna f\u00f6rb\u00e4ttra effektiviteten i cancerbehandlingen. Dessutom skulle o\u00f6nskade biverkningar ocks\u00e5 kunna minskas.<\/p>\n<p>F\u00f6r att dessa r\u00f6n snart ska kunna inf\u00f6rlivas i klinisk praxis m\u00e5ste resultaten valideras i studier med st\u00f6rre patientgrupper.
