Les <a href=\"https:\/\/www.nature.com\/articles\/s42003-024-06314-2\" target=\"_blank\" rel=\"noopener\">conclusions<\/a> des chercheurs, r\u00e9cemment publi\u00e9es dans la revue Nature Communications Biology, mettent en \u00e9vidence leur d\u00e9couverte des effets hypolipid\u00e9miants du peptide apparent\u00e9 \u00e0 la nesfatine-1 (NLP). Ce peptide &#8211; ou petite prot\u00e9ine &#8211; nouvellement identifi\u00e9 est un proche parent de la nesfatine-1 (NESF-1), qui r\u00e9gule la prise alimentaire et le poids corporel.<\/p>\n<p>La nesfatine-1 est un peptide s\u00e9cr\u00e9t\u00e9 par les tissus p\u00e9riph\u00e9riques et le syst\u00e8me nerveux central et p\u00e9riph\u00e9rique. Elle est impliqu\u00e9e dans la r\u00e9gulation de l&rsquo;hom\u00e9ostasie \u00e9nerg\u00e9tique en relation avec la r\u00e9gulation de la prise alimentaire et hydrique. La nesfatine-1 peut traverser la barri\u00e8re h\u00e9mato-enc\u00e9phalique dans les deux sens. Elle supprime la prise alimentaire ind\u00e9pendamment de la voie de signalisation de la leptine et augmente la s\u00e9cr\u00e9tion d&rsquo;insuline par les cellules des \u00eelots de Langerhans du pancr\u00e9as. Des chercheurs du WCVM et du USask College of Medicine ont particip\u00e9 \u00e0 cette \u00e9tude conjointe. Bien que l&rsquo;effet hypolipid\u00e9miant de la nesfatine-1 ait d\u00e9j\u00e0 \u00e9t\u00e9 signal\u00e9, Unniappan explique que l&rsquo;identification de la NLP et la compr\u00e9hension de ses capacit\u00e9s hypolipid\u00e9miantes dans les cellules humaines repr\u00e9sentent de nouvelles avanc\u00e9es dans le domaine de l&rsquo;endocrinologie.<\/p>\n<p>Toutefois, les chercheurs sont encore loin de pouvoir mettre ces r\u00e9sultats en pratique, mais ils disposent d\u00e9sormais de cibles multiples suppl\u00e9mentaires qui pourraient \u00eatre \u00e9tudi\u00e9es pour le traitement des troubles lipidiques et les progr\u00e8s th\u00e9rapeutiques. La d\u00e9couverte de l&rsquo;\u00e9quipe de recherche est une nouvelle encourageante \u00e9tant donn\u00e9 le manque de nouvelles th\u00e9rapies pour de nombreuses maladies m\u00e9taboliques &#8211; y compris la <a href=\"https:\/\/www.chronobiology.com\/non-alcoholic-fatty-liver-disease-can-damage-the-brain\/\" target=\"_blank\" rel=\"noopener\">st\u00e9atose h\u00e9patique non alcoolique<\/a> (r\u00e9cemment rebaptis\u00e9e maladie st\u00e9atosique du foie associ\u00e9e \u00e0 un dysfonctionnement m\u00e9tabolique ou MAFLD). Un m\u00e9dicament \u00e0 base d&rsquo;hormones a \u00e9t\u00e9 approuv\u00e9 aux \u00c9tats-Unis en mars 2024, mais il n&rsquo;existe \u00e0 ce jour aucun m\u00e9dicament destin\u00e9 exclusivement au traitement de cette maladie.<\/p>\n<h2>Une prot\u00e9ine sp\u00e9cifique abaisse les taux de lipides<\/h2>\n<p>Les plans de traitement typiques pour les personnes et les animaux souffrant de <a href=\"https:\/\/www.chronobiology.com\/the-role-of-the-circadian-clock-in-metabolic-diseases\/\" target=\"_blank\" rel=\"noopener\">maladies m\u00e9taboliques<\/a> consistent g\u00e9n\u00e9ralement en des changements de r\u00e9gime alimentaire et de l&rsquo;exercice physique pour diminuer progressivement le <a href=\"https:\/\/www.chronobiology.com\/stress-and-sleep-how-everyday-stressors-ruin-sleep-and-contribute-to-weight-gain\/\" target=\"_blank\" rel=\"noopener\">poids corporel<\/a> et r\u00e9duire l&rsquo;accumulation de graisse. Unniappan et son \u00e9quipe de recherche sont \u00e0 la pointe de la recherche sur la nesfatine-1. D\u00e9couverte en 2006 par un groupe de chercheurs japonais, la nesfatine-1 \u00e9tait initialement connue pour sa capacit\u00e9 \u00e0 supprimer la prise de nourriture.<\/p>\n<p>L&rsquo;\u00e9quipe d&rsquo;USask est all\u00e9e plus loin que les \u00e9tudes pr\u00e9c\u00e9dentes et a r\u00e9ussi \u00e0 d\u00e9montrer que la perturbation g\u00e9n\u00e9tique de la NLP entra\u00eene des changements dans les g\u00e8nes impliqu\u00e9s dans le m\u00e9tabolisme des graisses chez les souris. Ils ont constat\u00e9 que la perturbation du g\u00e8ne \u00e0 l&rsquo;origine de cette prot\u00e9ine [NLP], qui existe naturellement chez ces animaux, entra\u00eene en fait des modifications des g\u00e8nes li\u00e9s au m\u00e9tabolisme des graisses. La d\u00e9couverte de tels r\u00e9sultats &#8211; l&rsquo;administration de la NLP fait baisser les taux de lipides, tandis que la perturbation de sa production modifie le m\u00e9tabolisme des lipides &#8211; souligne son r\u00f4le central dans la r\u00e9gulation du m\u00e9tabolisme.<\/p>\n<p>Unniappan et Atefeh Nasri (PhD), qui a termin\u00e9 son programme de doctorat \u00e0 l&rsquo;USask en 2023 et est maintenant postdoctorante \u00e0 l&rsquo;Universit\u00e9 Dalhousie, ont collabor\u00e9 avec le Dr Scott Widenmaier (PhD), professeur adjoint d&rsquo;anatomie, de physiologie et de pharmacologie et expert en maladies m\u00e9taboliques au Coll\u00e8ge de m\u00e9decine de l&rsquo;USask. Le quatri\u00e8me membre de l&rsquo;\u00e9quipe \u00e9tait l&rsquo;\u00e9tudiant dipl\u00f4m\u00e9 Mateh Kowaluk.<\/p>\n<p>Unniappan esp\u00e8re que cette nouvelle recherche ouvrira la voie \u00e0 d&rsquo;autres recherches sur les options de traitement. Il pr\u00e9voit de travailler avec ses collaborateurs pour \u00e9tendre cette recherche \u00e0 des mod\u00e8les animaux plus complexes &#8211; y compris les rongeurs &#8211; et \u00e9ventuellement d&rsquo;\u00e9tudier des animaux plus grands tels que les chats et les chiens.
