De nombreuses personnes souffrent \u00e9galement de troubles cognitifs et fonctionnels. On sait que les patients atteints de schizophr\u00e9nie pr\u00e9sentent des perturbations de divers types de <a href=\"https:\/\/www.chronobiology.com\/non-24-hour-sleep-disorder\/\" target=\"_blank\" rel=\"noopener\">rythmes corporels sur 24 heures<\/a>, notamment les cycles veille-sommeil, les niveaux d&rsquo;hormones et l&rsquo;activit\u00e9 des g\u00e8nes dans le cortex pr\u00e9frontal du cerveau. Cependant, on ne sait pratiquement rien de l&rsquo;activit\u00e9 des g\u00e8nes dans le cerveau &#8211; sain ou non &#8211; pour des cycles plus courts que le rythme circadien habituel.<\/p>\n<h2>Activit\u00e9 g\u00e9n\u00e9tique cyclique anormale sur 12 heures dans le cerveau des personnes atteintes de schizophr\u00e9nie<\/h2>\n<p>Les niveaux de transcription des g\u00e8nes ne pouvant \u00eatre mesur\u00e9s dans les cerveaux vivants, la nouvelle <a href=\"https:\/\/journals.plos.org\/plosbiology\/article?id=10.1371\/journal.pbio.3001688\" target=\"_blank\" rel=\"noopener\">\u00e9tude<\/a> a utilis\u00e9 l&rsquo;analyse de l&rsquo;heure de la mort pour rechercher des <a href=\"https:\/\/www.chronobiology.com\/your-12-hour-internal-clock-how-ultradian-rhythms-affect-overall-health\/\" target=\"_blank\" rel=\"noopener\">rythmes de 12 heures<\/a> dans l&rsquo;activit\u00e9 des g\u00e8nes dans les cerveaux post-mortem. Les chercheurs se sont concentr\u00e9s sur le cortex pr\u00e9frontal dorsolat\u00e9ral, car cette r\u00e9gion du cerveau est associ\u00e9e \u00e0 des sympt\u00f4mes cognitifs et \u00e0 d&rsquo;autres anomalies dans les rythmes d&rsquo;expression des g\u00e8nes observ\u00e9s dans la schizophr\u00e9nie.<\/p>\n<p>Les chercheurs ont trouv\u00e9 de nombreux g\u00e8nes dans le cortex pr\u00e9frontal dorsolat\u00e9ral normal qui pr\u00e9sentent un rythme de 12 heures. Parmi eux, les niveaux d&rsquo;activit\u00e9 des g\u00e8nes li\u00e9s \u00e0 l&rsquo;\u00e9tablissement de connexions entre les <a href=\"https:\/\/www.chronobiology.com\/generation-of-new-neurons-in-the-brain-as-a-strategy-against-neurodegenerative-diseases\/\" target=\"_blank\" rel=\"noopener\">neurones<\/a> culminent respectivement l&rsquo;apr\u00e8s-midi et la nuit, tandis que ceux li\u00e9s \u00e0 la fonction mitochondriale (et donc \u00e0 l&rsquo;approvisionnement en \u00e9nergie cellulaire) culminent le matin et le soir.<\/p>\n<p>En revanche, les cerveaux post-mortem des patients schizophr\u00e8nes contenaient moins de g\u00e8nes avec des cycles d&rsquo;activit\u00e9 de 12 heures, et ceux impliqu\u00e9s dans les connexions neuronales \u00e9taient compl\u00e8tement absents. En outre, bien que les g\u00e8nes li\u00e9s \u00e0 la mitochondrie aient conserv\u00e9 un rythme de 12 heures, leur activit\u00e9 ne culminait pas aux heures normales. De futures \u00e9tudes devraient permettre de d\u00e9terminer si ces rythmes anormaux sont \u00e0 l&rsquo;origine des <a href=\"https:\/\/www.chronobiology.com\/lack-of-sleep-leads-to-negativity-changes-in-emotional-behavior\/\" target=\"_blank\" rel=\"noopener\">probl\u00e8mes comportementaux<\/a> de la schizophr\u00e9nie ou s&rsquo;ils sont dus aux m\u00e9dicaments, \u00e0 la consommation de nicotine ou aux troubles du sommeil.<\/p>\n<p>Les chercheurs notent que le cerveau humain pr\u00e9sente non seulement des <a href=\"https:\/\/www.chronobiology.com\/fighting-cancer-place-24-hour-rhythms\/\" target=\"_blank\" rel=\"noopener\">rythmes circadiens (24 heures)<\/a> dans l&rsquo;expression des g\u00e8nes, mais aussi des rythmes de 12 heures dans un certain nombre de g\u00e8nes importants pour la fonction cellulaire et le maintien des cellules nerveuses. Un grand nombre de ces rythmes d&rsquo;expression g\u00e9nique sont perdus chez les personnes atteintes de schizophr\u00e9nie, et l&rsquo;on observe un d\u00e9calage spectaculaire des rythmes dans les transcriptions mitochondriales, ce qui peut conduire \u00e0 une fonction mitochondriale sous-optimale aux moments de la journ\u00e9e o\u00f9 l&rsquo;\u00e9nergie cellulaire est n\u00e9cessaire.<\/p>\n<h2>Le risque de d\u00e9mence dans la schizophr\u00e9nie est fortement accru<\/h2>\n<p><a href=\"https:\/\/www.chronobiology.com\/wp-content\/uploads\/2023\/03\/shutterstock_1454425109.jpg\"><img decoding=\"async\" class=\"alignright wp-image-45576 size-medium\" title=\"shutterstock 1454425109\" src=\"https:\/\/www.chronobiology.com\/wp-content\/uploads\/2023\/03\/shutterstock_1454425109-300x169.jpg\" alt=\"shutterstock 1454425109\" width=\"300\" height=\"169\" srcset=\"https:\/\/dev.chronobiology.com\/wp-content\/uploads\/2023\/03\/shutterstock_1454425109-300x169.jpg 300w, https:\/\/dev.chronobiology.com\/wp-content\/uploads\/2023\/03\/shutterstock_1454425109-768x432.jpg 768w, https:\/\/dev.chronobiology.com\/wp-content\/uploads\/2023\/03\/shutterstock_1454425109-358x200.jpg 358w, https:\/\/dev.chronobiology.com\/wp-content\/uploads\/2023\/03\/shutterstock_1454425109.jpg 1000w\" sizes=\"(max-width: 300px) 100vw, 300px\" \/><\/a>Les personnes souffrant de troubles psychotiques tels que la schizophr\u00e9nie sont 2,5 fois plus susceptibles de d\u00e9velopper une <a href=\"https:\/\/www.chronobiology.com\/talking-sleep-may-indicate-greater-risk-developing-dementia\/\" target=\"_blank\" rel=\"noopener\">d\u00e9mence<\/a> que les personnes ne souffrant pas de troubles psychotiques, selon une analyse des donn\u00e9es probantes r\u00e9alis\u00e9e par des chercheurs de l&rsquo;UCL. L&rsquo;examen syst\u00e9matique et la m\u00e9ta-analyse, <a href=\"https:\/\/www.cambridge.org\/core\/journals\/psychological-medicine\/article\/nonaffective-psychotic-disorders-and-risk-of-dementia-a-systematic-review-and-metaanalysis\/7D08C3689D40151B5B55EC639A325F98\" target=\"_blank\" rel=\"noopener\">publi\u00e9s dans Psychological Medicine<\/a>, ont r\u00e9v\u00e9l\u00e9 que les troubles psychotiques peuvent avoir une association plus forte avec la d\u00e9mence que d&rsquo;autres troubles mentaux tels que la d\u00e9pression ou l&rsquo;anxi\u00e9t\u00e9.<\/p>\n<p>Les chercheurs ont rassembl\u00e9 des donn\u00e9es provenant de 11 \u00e9tudes r\u00e9alis\u00e9es dans neuf pays sur quatre continents, soit pr\u00e8s de 13 millions de participants. Ils ont constat\u00e9 que, pour toute une s\u00e9rie de <a href=\"https:\/\/www.chronobiology.com\/role-melatonin-variety-neuropsychiatric-disorders\/\" target=\"_blank\" rel=\"noopener\">troubles psychotiques<\/a>, et quel que soit l&rsquo;\u00e2ge auquel la maladie mentale s&rsquo;est d\u00e9clar\u00e9e, le risque de d\u00e9mence est plus \u00e9lev\u00e9 \u00e0 un stade ult\u00e9rieur de la vie. Certaines \u00e9tudes ont port\u00e9 sur des personnes chez qui des troubles psychotiques avaient \u00e9t\u00e9 diagnostiqu\u00e9s au d\u00e9but de l&rsquo;\u00e2ge adulte et ont \u00e9t\u00e9 suivies pendant plusieurs dizaines d&rsquo;ann\u00e9es. Elles ont \u00e9galement montr\u00e9 que les personnes souffrant de troubles psychotiques avaient tendance \u00e0 \u00eatre plus jeunes que la moyenne lorsqu&rsquo;on leur diagnostiquait une d\u00e9mence, et deux \u00e9tudes ont montr\u00e9 que les personnes souffrant de troubles psychotiques \u00e9taient beaucoup plus susceptibles de recevoir un diagnostic de d\u00e9mence alors qu&rsquo;elles avaient encore la soixantaine.<\/p>\n<h3>Les causes ne sont pas claires<\/h3>\n<p>Des recherches ant\u00e9rieures ont montr\u00e9 que la <a href=\"https:\/\/www.chronobiology.com\/new-link-found-between-sleep-apnea-and-depression\/\" target=\"_blank\" rel=\"noopener\">d\u00e9pression<\/a> et l&rsquo;anxi\u00e9t\u00e9 augmentaient \u00e9galement le risque de d\u00e9mence, mais des r\u00e9sultats r\u00e9cents sugg\u00e8rent que les troubles psychotiques sont plus fortement associ\u00e9s au risque de d\u00e9mence. Les chercheurs n&rsquo;ont pas pu confirmer la cause de cette association, qu&rsquo;elle soit due \u00e0 la maladie mentale elle-m\u00eame ou peut-\u00eatre au fait que les troubles psychotiques augmentent la probabilit\u00e9 de maladies qui, \u00e0 leur tour, augmentent le risque de d\u00e9mence.<\/p>\n<p>Le lien peut s&rsquo;expliquer en partie par le fait que les sympt\u00f4mes psychotiques peuvent \u00eatre des signes pr\u00e9coces de d\u00e9mence chez certaines personnes, mais le fait que certaines des \u00e9tudes comportaient des p\u00e9riodes de suivi tr\u00e8s longues et incluaient des personnes ayant souffert de psychose \u00e0 un jeune \u00e2ge sugg\u00e8re que ce n&rsquo;est pas la seule explication. Selon les experts, les personnes souffrant de troubles psychotiques peuvent \u00eatre plus susceptibles d&rsquo;avoir d&rsquo;autres probl\u00e8mes de sant\u00e9, tels que les maladies cardiovasculaires ou l&rsquo;<a href=\"https:\/\/www.chronobiology.com\/scientists-discover-why-eating-late-increases-obesity-risk\/\" target=\"_blank\" rel=\"noopener\">ob\u00e9sit\u00e9<\/a>.
