Cette d\u00e9couverte permet d&rsquo;expliquer pourquoi les modifications du rythme circadien de l&rsquo;organisme, telles que le travail de nuit et le d\u00e9calage horaire, peuvent augmenter le risque de maladies m\u00e9taboliques telles que le <a href=\"https:\/\/www.chronobiology.com\/circadian-rhythm-and-diabetes-risk-whats-the-connection\/\" target=\"_blank\" rel=\"noopener\">diab\u00e8te<\/a>.<\/p>\n<h2>Comment la privation de glucose affecte l&rsquo;horloge circadienne<\/h2>\n<p>L&rsquo;horloge circadienne est \u00e9troitement li\u00e9e au m\u00e9tabolisme : d&rsquo;une part, elle module rythmiquement de nombreuses voies m\u00e9taboliques, d&rsquo;autre part, les nutriments et les stimuli m\u00e9taboliques influencent la fonction de l&rsquo;horloge. Cela se fait par le biais de boucles de r\u00e9troaction finement r\u00e9gl\u00e9es, o\u00f9 certains composants positifs de l&rsquo;horloge en activent d&rsquo;autres, qui r\u00e9troagissent n\u00e9gativement sur les composants activateurs d&rsquo;origine. Comme le glucose affecte de nombreuses voies de signalisation, on pense qu&rsquo;un manque de glucose pourrait remettre en question les boucles de r\u00e9troaction de l&rsquo;horloge circadienne et entraver sa capacit\u00e9 \u00e0 maintenir un rythme constant. Ils ont constat\u00e9 que les niveaux de WC1 et 2 diminuaient progressivement pour atteindre respectivement environ 15 % et 20 % des niveaux initiaux avant la famine, tandis que les niveaux de FRQ restaient inchang\u00e9s mais \u00e9taient modifi\u00e9s par l&rsquo;ajout de nombreux groupes phosphates (un processus connu sous le nom d&rsquo;hyperphosphorylation, ou hyperphosphorylation). Normalement, l&rsquo;hyperphosphorylation emp\u00eache FRQ d&rsquo;inhiber l&rsquo;activit\u00e9 du CMC, c&rsquo;est pourquoi les auteurs ont suppos\u00e9 que l&rsquo;activit\u00e9 plus \u00e9lev\u00e9e pouvait acc\u00e9l\u00e9rer la d\u00e9gradation du CMC. Dans l&rsquo;ensemble, ces r\u00e9sultats sugg\u00e8rent que l&rsquo;<a href=\"https:\/\/www.chronobiology.com\/new-discoveries-in-chronobiology-the-circadian-clock-protects-eyes-as-they-age\/\" target=\"_blank\" rel=\"noopener\"> horloge circadienne<\/a> fonctionne toujours correctement, entra\u00eenant l&rsquo;expression rythmique des g\u00e8nes cellulaires pendant la privation de glucose.<\/p>\n<p>Pour approfondir l&rsquo;\u00e9tude de l&rsquo;importance de l&rsquo;horloge mol\u00e9culaire dans l&rsquo;adaptation \u00e0 la privation de glucose, l&rsquo;\u00e9quipe a utilis\u00e9 une souche de Neurospora d\u00e9pourvue du domaine WC-1 du WCC. Ils ont ensuite compar\u00e9 le niveau d&rsquo;expression des g\u00e8nes apr\u00e8s une privation de glucose avec celui d&rsquo;une souche de Neurospora dont l&rsquo;horloge mol\u00e9culaire \u00e9tait intacte. Ils ont constat\u00e9 que la privation de <a href=\"https:\/\/www.chronobiology.com\/blood-glucose-and-your-internal-clock\/\" target=\"_blank\" rel=\"noopener\">glucose<\/a> \u00e0 long terme affectait plus de 20 % des g\u00e8nes codants et que 1 377 de ces 9 758 g\u00e8nes codants (13 %) pr\u00e9sentaient des changements sp\u00e9cifiques \u00e0 la souche, selon que les cellules poss\u00e9daient ou non une horloge mol\u00e9culaire. Cela signifie que l&rsquo;horloge est un \u00e9l\u00e9ment important de la machinerie qui permet aux cellules de r\u00e9agir \u00e0 un manque de glucose.<\/p>\n<h2>L&rsquo;horloge interne r\u00e9gule le m\u00e9tabolisme et la sant\u00e9<\/h2>\n<p>L&rsquo;\u00e9quipe a ensuite cherch\u00e9 \u00e0 savoir si une horloge fonctionnelle \u00e9tait importante pour que les cellules se r\u00e9tablissent apr\u00e8s une privation de glucose. Elle a constat\u00e9 que les cellules de Neurospora d\u00e9pourvues d&rsquo;un FRQ ou d&rsquo;un WCC fonctionnel se d\u00e9veloppaient beaucoup plus lentement que les cellules normales lorsque du glucose \u00e9tait ajout\u00e9, ce qui sugg\u00e8re qu&rsquo;une horloge fonctionnelle aide les <a href=\"https:\/\/www.chronobiology.com\/the-connection-between-stem-cell-circadian-rhythm-and-aging\/\" target=\"_blank\" rel=\"noopener\">cellules<\/a> \u00e0 se r\u00e9g\u00e9n\u00e9rer.
