Nous savons que les rythmes circadiens sont g\u00e9n\u00e9r\u00e9s par des \u00ab\u00a0g\u00e8nes d&rsquo;horloge\u00a0\u00bb qui affectent la plupart des organes et des cellules, y compris celles du syst\u00e8me immunitaire, dont la fonction varie en fonction de l&rsquo;heure de la journ\u00e9e. En cons\u00e9quence, on trouve des <a href=\"https:\/\/www.chronobiology.com\/researchers-investigate-tracking-circadian-rhythms-with-your-smart-watch\/\" target=\"_blank\" rel=\"noopener\">rythmes circadiens<\/a> pour divers aspects de la physiologie, notamment le sommeil, la nutrition, l&rsquo;activit\u00e9 hormonale et la temp\u00e9rature corporelle. Ces rythmes quotidiens aident l&rsquo;organisme \u00e0 s&rsquo;adapter aux changements cycliques de l&rsquo;environnement, tels que les saisons et le cycle jour-nuit.<\/p>\n<h2>Rythme circadien et heure de la vaccination<\/h2>\n<p>Des recherches men\u00e9es par l&rsquo;Universit\u00e9 de m\u00e9decine et des sciences de la sant\u00e9 RCSI ont permis de mieux comprendre le m\u00e9canisme par lequel notre horloge circadienne de 24 heures affecte notre r\u00e9ponse immunitaire aux vaccins en fonction de l&rsquo;heure de la journ\u00e9e. L&rsquo;\u00e9tude a examin\u00e9 les changements qui se produisent dans les <a href=\"https:\/\/www.chronobiology.com\/circadian-rhythms-and-mitochondria-the-chronobiology-of-energy-production\/\" target=\"_blank\" rel=\"noopener\">mitochondries<\/a> d&rsquo;une cellule immunitaire cl\u00e9 impliqu\u00e9e dans la r\u00e9ponse vaccinale et pourrait contribuer \u00e0 am\u00e9liorer la conception et le calendrier des futurs vaccins afin d&rsquo;en maximiser l&rsquo;efficacit\u00e9.<\/p>\n<p>Il avait d\u00e9j\u00e0 \u00e9t\u00e9 constat\u00e9 que les personnes r\u00e9agissaient plus fortement \u00e0 certains vaccins en fonction du moment de la journ\u00e9e o\u00f9 ils \u00e9taient administr\u00e9s, mais la raison de ce ph\u00e9nom\u00e8ne n&rsquo;\u00e9tait pas tout \u00e0 fait claire. Les scientifiques ont maintenant d\u00e9couvert <a href=\"https:\/\/www.nature.com\/articles\/s41467-022-34897-z\" target=\"_blank\" rel=\"noopener\">que notre horloge circadienne modifie la forme des mitochondries dans les cellules dendritiques<\/a>. Dans la formation en r\u00e9seau, la vaccination est plus efficace car les cellules dendritiques sont mieux \u00e0 m\u00eame de d\u00e9composer le vaccin en petits morceaux pour interagir avec nos cellules immunitaires (cellules T). Dans l&rsquo;\u00e9tude, les chercheurs ont utilis\u00e9 une approche pour induire la phase r\u00e9ticul\u00e9e, ce qui pourrait avoir des implications pour la conception des vaccins et nous permettre d&rsquo;optimiser notre <a href=\"https:\/\/www.chronobiology.com\/how-time-of-day-affects-immune-system-function\/\" target=\"_blank\" rel=\"noopener\">r\u00e9ponse immunitaire<\/a> quelle que soit l&rsquo;heure de la journ\u00e9e. Cette compr\u00e9hension pourrait \u00eatre utile pour s&rsquo;assurer que nous tirons le maximum de b\u00e9n\u00e9fices de la vaccination.<\/p>\n<h2>Association entre la r\u00e9ponse des anticorps au vaccin COVID-19 et le moment de la journ\u00e9e<\/h2>\n<p>Quel r\u00f4le joue le moment de la journ\u00e9e dans la vaccination contre le SRAS-CoV-2 ? Une <a href=\"https:\/\/journals.sagepub.com\/doi\/10.1177\/07487304221124661\" target=\"_blank\" rel=\"noopener\">\u00e9tude men\u00e9e au Japon a montr\u00e9<\/a> que la r\u00e9ponse en anticorps au vaccin \u00e0 ARNm COVID-19 de Moderna ne d\u00e9pendait pas de l&rsquo;heure \u00e0 laquelle la dose \u00e9tait administr\u00e9e. Plusieurs \u00e9tudes ont examin\u00e9 si la r\u00e9ponse immunitaire aux vaccins COVID-19 variait en fonction du moment de la journ\u00e9e o\u00f9 la dose \u00e9tait administr\u00e9e, mais les r\u00e9sultats d\u00e9pendaient du type de vaccin, de l&rsquo;\u00e2ge et du sexe des participants. Des scientifiques de l&rsquo;universit\u00e9 d&rsquo;Hokkaido et des coll\u00e8gues au Japon ont men\u00e9 une \u00e9tude qui n&rsquo;a trouv\u00e9 aucun lien entre la r\u00e9ponse des anticorps au vaccin COVID-19 et le moment de la journ\u00e9e o\u00f9 la vaccination a \u00e9t\u00e9 effectu\u00e9e.<\/p>\n<p>Les chercheurs ont recrut\u00e9 les participants \u00e0 leur \u00e9tude parmi le personnel et les \u00e9tudiants de l&rsquo;universit\u00e9 d&rsquo;Hokkaido qui avaient re\u00e7u leur premi\u00e8re dose du vaccin mRNA-1273 (Moderna) deux \u00e0 quatre semaines auparavant. Les personnes pr\u00e9c\u00e9demment infect\u00e9es par le <a href=\"https:\/\/www.chronobiology.com\/sleep-the-circadian-rhythm-and-covid-19\/\" target=\"_blank\" rel=\"noopener\">COVID-19<\/a> ont \u00e9t\u00e9 exclues. Le groupe d&rsquo;\u00e9tude n&rsquo;incluait pas les travailleurs post\u00e9s, ce qui constitue une diff\u00e9rence essentielle entre cette \u00e9tude et d&rsquo;autres \u00e9tudes ant\u00e9rieures portant sur des travailleurs m\u00e9dicaux.<\/p>\n<p>L&rsquo;\u00e9quipe a recueilli les donn\u00e9es d\u00e9mographiques du dernier groupe de 332 participants japonais et a pr\u00e9lev\u00e9 des \u00e9chantillons de sang pour mesurer les titres d&rsquo;anticorps contre le SRAS-CoV-2. Ils ont ensuite effectu\u00e9 des analyses statistiques pour v\u00e9rifier s&rsquo;il existait des liens entre l&rsquo;heure \u00e0 laquelle les participants ont re\u00e7u leur dose de vaccin, l&rsquo;une ou l&rsquo;autre des caract\u00e9ristiques d\u00e9mographiques et la r\u00e9ponse immunitaire.<\/p>\n<p>Leurs analyses n&rsquo;ont pas r\u00e9v\u00e9l\u00e9 de lien significatif entre l&rsquo;heure de la vaccination et la concentration d&rsquo;anticorps contre le SRAS-CoV-2, mais elles ont montr\u00e9 que les titres d&rsquo;anticorps diminuaient chez les participants \u00e2g\u00e9s de 50 \u00e0 64 ans, ce qui est conforme aux \u00e9tudes pr\u00e9c\u00e9dentes. Toutefois, les chercheurs admettent que d&rsquo;autres \u00e9tudes sont n\u00e9cessaires pour d\u00e9terminer si et dans quelle mesure les rythmes circadiens influencent la r\u00e9ponse des anticorps \u00e0 la vaccination.<\/p>\n<h2>Les cellules T r\u00e9agissent diff\u00e9remment aux corps \u00e9trangers \u00e0 certains moments de la journ\u00e9e<\/h2>\n<p>Selon une \u00e9tude publi\u00e9e dans les Proceedings of the National Academy of Sciences, l&rsquo;<a href=\"https:\/\/www.pnas.org\/doi\/full\/10.1073\/pnas.1905080116\" target=\"_blank\" rel=\"noopener\">horloge biologique influence l&rsquo;efficacit\u00e9 de la r\u00e9ponse immunitaire<\/a>. En effet, les cellules T CD8, essentielles pour lutter contre les infections et le cancer, fonctionnent tr\u00e8s diff\u00e9remment selon l&rsquo;heure de la journ\u00e9e. L&rsquo;\u00e9tude a \u00e9t\u00e9 men\u00e9e par une \u00e9quipe de chercheurs dirig\u00e9e par Nicolas Cermakian, Ph.D., du Centre de recherche du Douglas, et Nathalie Labrecque, Ph.D., du Centre de recherche de l&rsquo;H\u00f4pital Maisonneuve-Rosemont.<\/p>\n<p>Dans des recherches ant\u00e9rieures, l&rsquo;\u00e9quipe avait d\u00e9montr\u00e9 que les <a href=\"https:\/\/www.chronobiology.com\/researchers-are-discovering-new-ways-to-improve-cancer-therapies\/\" target=\"_blank\" rel=\"noopener\">cellules T<\/a> r\u00e9pondent plus ou moins fortement \u00e0 un corps \u00e9tranger selon l&rsquo;heure du jour, mais le r\u00f4le de l&rsquo;horloge biologique dans ce ph\u00e9nom\u00e8ne demeurait inconnu. En utilisant un mod\u00e8le de vaccination chez la souris, les scientifiques ont d\u00e9couvert que l&rsquo;intensit\u00e9 de la r\u00e9ponse des cellules T CD8 apr\u00e8s la vaccination variait en fonction de l&rsquo;heure de la journ\u00e9e. \u00c0 l&rsquo;inverse, chez les souris dont les cellules T CD8 pour le g\u00e8ne de l&rsquo;horloge \u00e9taient d\u00e9ficientes, ce rythme circadien \u00e9tait aboli et la r\u00e9ponse au vaccin \u00e9tait plus faible pendant la journ\u00e9e.<\/p>\n<p>Les r\u00e9sultats de la recherche montrent que les cellules T sont plus enclines \u00e0 \u00eatre activ\u00e9es \u00e0 certains moments de la journ\u00e9e.
