Des sch\u00e9mas th\u00e9rapeutiques individualis\u00e9s pourraient se traduire par des traitements <a href=\"https:\/\/bmccancer.biomedcentral.com\/articles\/10.1186\/s12885-017-3896-y\" target=\"_blank\" rel=\"noopener noreferrer\">plus efficaces et moins toxiques<\/a>.<\/p>\n<h2>Que sont les rythmes circadiens ?<\/h2>\n<p>Tous les \u00eatres vivants expos\u00e9s \u00e0 la lumi\u00e8re du soleil poss\u00e8dent une horloge biologique dont les rythmes inn\u00e9s tendent \u00e0 s&rsquo;aligner sur leur environnement. Les <a href=\"https:\/\/www.chronobiology.com\/why-you-should-maintain-a-healthy-circadian-rhythm-in-2018\/\" target=\"_blank\" rel=\"noopener noreferrer\">rythmes circadiens<\/a> sont le plus souvent associ\u00e9s \u00e0 leur r\u00f4le dans notre cycle veille-sommeil, mais la recherche continue \u00e0 d\u00e9couvrir \u00e0 quel point les nombreuses horloges du corps affectent notre fonctionnement biochimique, physiologique et psychologique.<\/p>\n<p>Chaque cellule de notre corps poss\u00e8de sa propre horloge. Ces horloges internes suivent un cycle de 24 heures et exercent une influence sur tout, de la faim \u00e0 la clart\u00e9 cognitive, en passant par la fonction cardiaque, l&rsquo;immunit\u00e9, l&rsquo;humeur et les m\u00e9canismes de r\u00e9ponse au stress. Des preuves de plus en plus nombreuses \u00e9tablissent un lien entre le d\u00e9r\u00e8glement de l&rsquo;horloge biologique et de nombreux probl\u00e8mes de sant\u00e9. Il est donc important de mieux comprendre la relation entre notre corps et ses rythmes naturels.<\/p>\n<h2>Chronobiologie et chimioth\u00e9rapie : Les rythmes circadiens influencent les traitements contre le cancer<\/h2>\n<p>Un groupe de chercheurs du <a href=\"http:\/\/sancarlab.unc.edu\/\" target=\"_blank\" rel=\"noopener noreferrer\">laboratoire Sancar<\/a> de l&rsquo;universit\u00e9 de Caroline du Nord \u00e0 Chapel Hill s&rsquo;est int\u00e9ress\u00e9 \u00e0 l&rsquo;impact direct de ces rythmes circadiens sur la r\u00e9paration de l&rsquo;ADN dans les cellules. Les <a href=\"https:\/\/pubs.acs.org\/doi\/10.1021\/bi5007354\" target=\"_blank\" rel=\"noopener noreferrer\">recherches de<\/a> l&rsquo;\u00e9quipe concernant la chimioth\u00e9rapie et les dommages cellulaires sugg\u00e8rent que pour obtenir les meilleurs r\u00e9sultats avec le moins d&rsquo;effets secondaires possible, il existe un moment id\u00e9al pour administrer les m\u00e9dicaments chimioth\u00e9rapeutiques.<\/p>\n<p>Selon Aziz Sancar, professeur de biochimie et de biophysique \u00e0 l&rsquo;universit\u00e9 de Caroline du Nord \u00e0 Chapel Hill, \u00ab\u00a0nous avons d\u00e9couvert qu&rsquo;il existe pr\u00e8s de 2 000 g\u00e8nes, dont diff\u00e9rentes parties sont r\u00e9par\u00e9es \u00e0 diff\u00e9rents moments de la journ\u00e9e, en fonction du g\u00e8ne\u00a0\u00bb. Le professeur Sancar a re\u00e7u le prix Nobel de chimie en 2015 aux c\u00f4t\u00e9s de ses coll\u00e8gues pour leurs travaux sur la compr\u00e9hension des m\u00e9canismes sous-jacents de la r\u00e9paration de l&rsquo;ADN. L&rsquo;\u00e9quipe du laboratoire Sancar continue d&rsquo;approfondir ses recherches sur l&rsquo;impact des rythmes circadiens sur la r\u00e9paration de l&rsquo;ADN.<\/p>\n<h2>Cancer, cisplatine et r\u00f4le des rythmes circadiens dans le traitement<\/h2>\n<p>S&rsquo;appuyant sur leurs <a href=\"http:\/\/www.pnas.org\/content\/113\/41\/11507\" target=\"_blank\" rel=\"noopener noreferrer\">recherches ant\u00e9rieures<\/a> concernant l&rsquo;horloge circadienne et la chimioth\u00e9rapie, les nouveaux travaux de l&rsquo;\u00e9quipe permettent de mieux comprendre l&rsquo;impact des <a href=\"https:\/\/www.chronobiology.com\/new-research-suggests-circadian-clock-suppresses-cancer\/\" target=\"_blank\" rel=\"noopener noreferrer\">horloges biologiques<\/a> sur la r\u00e9paration de l&rsquo;ADN au cours des traitements de chimioth\u00e9rapie avec le cisplatine, un m\u00e9dicament anticanc\u00e9reux tr\u00e8s r\u00e9pandu. Dans ses travaux les plus r\u00e9cents, l&rsquo;\u00e9quipe a examin\u00e9 comment les g\u00e8nes des tissus r\u00e9naux et h\u00e9patiques des souris r\u00e9agissaient pendant les 24 heures qui suivaient le traitement au cisplatine. Leurs conclusions ont \u00e9t\u00e9 publi\u00e9es r\u00e9cemment dans la revue <a href=\"http:\/\/www.pnas.org\/content\/115\/21\/E4777\" target=\"_blank\" rel=\"noopener noreferrer\"><em>PNAS<\/em><\/a>.<\/p>\n<p>Le cisplatine est le m\u00e9dicament anticanc\u00e9reux le plus couramment utilis\u00e9 dans le traitement des cancers des tissus solides tels que les cancers du sein, du c\u00f4lon, du poumon, des ovaires et des testicules. Le m\u00e9dicament tue efficacement le cancer en endommageant son ADN cellulaire, mais il endommage \u00e9galement l&rsquo;ADN des cellules saines, cr\u00e9ant des effets secondaires nocifs qui n\u00e9cessitent parfois l&rsquo;arr\u00eat du traitement. \u00c9tant donn\u00e9 que les cellules saines et canc\u00e9reuses sont capables de r\u00e9parer les dommages cellulaires caus\u00e9s par le cisplatine, le succ\u00e8s du traitement repose sur l&rsquo;endommagement du tissu canc\u00e9reux lorsqu&rsquo;il est moins capable de se r\u00e9parer, tout en \u00e9pargnant autant que possible le tissu normal.<\/p>\n<p><a href=\"https:\/\/www.chronobiology.com\/wp-content\/uploads\/2018\/06\/chronobiology-and-chemotherapy-optimal-drug-timing-improves-treatment-effectiveness-2.jpg\"><img decoding=\"async\" class=\"size-medium wp-image-21990 alignright\" src=\"https:\/\/www.chronobiology.com\/wp-content\/uploads\/2018\/06\/chronobiology-and-chemotherapy-optimal-drug-timing-improves-treatment-effectiveness-2-300x199.jpg\" alt=\"Chronobiology and Chemotherapy: Optimal Drug Timing Improves Treatment Effectiveness\" width=\"300\" height=\"199\" srcset=\"https:\/\/dev.chronobiology.com\/wp-content\/uploads\/2018\/06\/chronobiology-and-chemotherapy-optimal-drug-timing-improves-treatment-effectiveness-2-300x200.jpg 300w, https:\/\/dev.chronobiology.com\/wp-content\/uploads\/2018\/06\/chronobiology-and-chemotherapy-optimal-drug-timing-improves-treatment-effectiveness-2-768x509.jpg 768w, https:\/\/dev.chronobiology.com\/wp-content\/uploads\/2018\/06\/chronobiology-and-chemotherapy-optimal-drug-timing-improves-treatment-effectiveness-2.jpg 800w\" sizes=\"(max-width: 300px) 100vw, 300px\" \/><\/a>Au cours des derni\u00e8res d\u00e9cennies, la <a href=\"http:\/\/doi.org\/10.1200\/JCO.2006.06.1440\" target=\"_blank\" rel=\"noopener noreferrer\">recherche<\/a> a examin\u00e9 le lien entre la chronobiologie et la chimioth\u00e9rapie dans l&rsquo;espoir de mieux guider le choix du moment de la th\u00e9rapie, mais si les <a href=\"http:\/\/doi.org\/10.1200\/JCO.1990.8.4.705\" target=\"_blank\" rel=\"noopener noreferrer\">r\u00e9sultats<\/a> \u00e9taient toujours prometteurs, leurs <a href=\"http:\/\/doi.org\/10.1200\/JCO.2003.10.083\" target=\"_blank\" rel=\"noopener noreferrer\">exp\u00e9riences<\/a> ont finalement \u00e9chou\u00e9 en raison de choix de temps arbitraires plut\u00f4t que de signaux de l&rsquo;horloge cellulaire. L&rsquo;\u00e9quipe du professeur Sancar a mis au point une technique permettant de contr\u00f4ler \u00e0 la fois comment et quand des segments pr\u00e9cis d&rsquo;ADN ont \u00e9t\u00e9 r\u00e9par\u00e9s. Dans ses travaux les plus r\u00e9cents, l&rsquo;\u00e9quipe a examin\u00e9 plus de 25 000 g\u00e8nes \u00e0 l&rsquo;aide de cette m\u00e9thode pour comprendre exactement comment l&rsquo;ADN des cellules r\u00e9nales et h\u00e9patiques de souris r\u00e9agissait apr\u00e8s avoir \u00e9t\u00e9 trait\u00e9 au cisplatine. Ils ont d\u00e9couvert que certains <a href=\"https:\/\/www.chronobiology.com\/new-research-cancer-genes-dictate-circadian-rhythm\/\" target=\"_blank\" rel=\"noopener noreferrer\">g\u00e8nes<\/a> se r\u00e9paraient toujours le matin, tandis que d&rsquo;autres se r\u00e9paraient toujours le soir.<\/p>\n<h2>Des g\u00e8nes diff\u00e9rents se r\u00e9parent \u00e0 des moments diff\u00e9rents de la journ\u00e9e<\/h2>\n<p><a href=\"https:\/\/theconversation.com\/chemotherapy-timing-could-influence-how-well-the-treatment-works-96258\" target=\"_blank\" rel=\"noopener noreferrer\">Selon Yanyan Yang<\/a>, chercheur postdoctoral en biochimie et biophysique \u00e0 l&rsquo;universit\u00e9 de Caroline du Nord \u00e0 Chapel Hill et principal chercheur de l&rsquo;\u00e9quipe, \u00ab\u00a0nous avons d\u00e9couvert quel g\u00e8ne causant le cancer, ou quel g\u00e8ne pr\u00e9venant le cancer, est r\u00e9par\u00e9 lorsque le m\u00e9dicament est administr\u00e9 \u00e0 certains moments de la journ\u00e9e. Certains g\u00e8nes sont toujours r\u00e9par\u00e9s \u00e0 l&rsquo;aube, d&rsquo;autres au cr\u00e9puscule. Cela sugg\u00e8re que le meilleur moment pour la chimioth\u00e9rapie est celui o\u00f9 les cellules saines peuvent reconstruire efficacement leur ADN et o\u00f9 les cellules tumorales se r\u00e9parent mal\u00a0\u00bb.<\/p>\n<p>L&rsquo;\u00e9quipe esp\u00e8re appliquer ses conclusions aux tissus humains dans de futures \u00e9tudes.
