Cependant, la force d&rsquo;un signal mol\u00e9culaire est \u00e9galement importante. Si un signal est trop faible, il n&rsquo;aura aucun effet. S&rsquo;il est trop fort, il risque d&rsquo;interf\u00e9rer avec d&rsquo;autres fonctions. En biologie mol\u00e9culaire comme en Boucles d&rsquo;or, seul le \u00ab\u00a0juste ce qu&rsquo;il faut\u00a0\u00bb fait l&rsquo;affaire. Les chercheurs ont d\u00e9couvert qu&rsquo;une prot\u00e9ine appel\u00e9e REV-ERB\u03b1, dont on sait d\u00e9j\u00e0 qu&rsquo;elle joue un r\u00f4le important dans le rythme circadien, pourrait \u00eatre le facteur qui contr\u00f4le l&rsquo;intensit\u00e9 du rythme circadien, en le maintenant dans la plage d&rsquo;amplitude o\u00f9 il est le plus efficace.<\/p>\n<h2>Qu&rsquo;est-ce que REV-ERB\u03b1 ?<\/h2>\n<p>Comme de nombreuses prot\u00e9ines du corps humain, REV-ERB\u03b1 semble jouer plusieurs r\u00f4les. Il fait partie int\u00e9grante des <a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/1971514\" target=\"_blank\" rel=\"noopener\">r\u00e9cepteurs des hormones thyro\u00efdiennes<\/a>, joue un r\u00f4le important dans la <a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/17075855\" target=\"_blank\" rel=\"noopener\">r\u00e9gulation du m\u00e9tabolisme du cartilage<\/a> et joue \u00e9galement un r\u00f4le important dans le rythme circadien. Son <a href=\"http:\/\/www.ncbi.nlm.nih.gov\/pubmed\/22460952\" target=\"_blank\" rel=\"noopener\">r\u00f4le dans le rythme circadien<\/a> a fait l&rsquo;objet de nombreuses recherches en raison du mode d&rsquo;action inhabituel de cette prot\u00e9ine. Au moment de la journ\u00e9e o\u00f9 certains g\u00e8nes circadiens ne doivent pas \u00eatre exprim\u00e9s, REV-ERB\u03b1 se lie \u00e0 l&rsquo;ADN de ces g\u00e8nes pour en arr\u00eater physiquement la transcription.<\/p>\n<h2>Le contr\u00f4le du volume de la force du rythme circadien<\/h2>\n<p>Comme de nombreuses prot\u00e9ines li\u00e9es au rythme circadien, REV-ERB\u03b1 est exprim\u00e9e de mani\u00e8re rythmique. Son niveau d&rsquo;expression est le plus \u00e9lev\u00e9 pendant la journ\u00e9e et atteint son niveau le plus bas pendant la nuit. Les chercheurs ont d\u00e9cid\u00e9 de d\u00e9sactiver l&rsquo;enzyme qui d\u00e9grade REV-ERB\u03b1, afin de maintenir ses niveaux \u00e9lev\u00e9s la nuit. Le rythme circadien n&rsquo;a pas chang\u00e9, mais les fluctuations de l&rsquo;expression des g\u00e8nes ont \u00e9t\u00e9 modifi\u00e9es. Les g\u00e8nes qui sont normalement exprim\u00e9s \u00e0 des niveaux variables tout au long d&rsquo;un cycle de 24 heures ont \u00e9t\u00e9 exprim\u00e9s \u00e0 des niveaux \u00e9lev\u00e9s quelle que soit l&rsquo;heure. En d&rsquo;autres termes, le rythme circadien est rest\u00e9 le m\u00eame, mais son volume \u00e9tait constant et \u00ab\u00a0fort\u00a0\u00bb au lieu d&rsquo;augmenter et de diminuer de mani\u00e8re rythmique.<\/p>\n<p>Les chercheurs pensent que REV-ERB\u03b1 pourrait \u00eatre le bouton de volume principal qui r\u00e9git l&rsquo;intensit\u00e9 du rythme circadien. L&rsquo;auteur principal, Ronald Evans, qui est \u00e9galement directeur du laboratoire d&rsquo;expression g\u00e9n\u00e9tique de Salk, a comment\u00e9 l&rsquo;\u00e9tude,<\/p>\n<blockquote>\n<p>\u00ab\u00a0Qu&rsquo;il s&rsquo;agisse de la 9e symphonie de Beethoven sur votre cha\u00eene st\u00e9r\u00e9o ou de la symphonie des g\u00e8nes dans notre corps, toutes deux ont besoin de volume pour \u00eatre entendues. Nos travaux r\u00e9cents d\u00e9crivent comment REV-ERB\u03b1 agit comme un chef d&rsquo;orchestre mol\u00e9culaire qui permet d&rsquo;augmenter ou de diminuer le volume ou l&rsquo;activit\u00e9 de milliers de g\u00e8nes\u00a0\u00bb.<\/p>\n<\/blockquote>\n<h2>L&rsquo;amplitude est essentielle<\/h2>\n<p>C&rsquo;est la premi\u00e8re fois que des scientifiques parviennent \u00e0 influencer exp\u00e9rimentalement la force du rythme circadien, mais il semble qu&rsquo;il s&rsquo;agisse d&rsquo;un facteur tr\u00e8s important. Dans cette \u00e9tude chronobiologique, des souris g\u00e9n\u00e9tiquement modifi\u00e9es pour manquer de REV-ERB\u03b1 ont d\u00e9velopp\u00e9 une s\u00e9rie de probl\u00e8mes de sant\u00e9 graves, notamment une st\u00e9atose h\u00e9patique. Une trop grande quantit\u00e9 de cette prot\u00e9ine pourrait \u00e9galement avoir de graves effets sur la sant\u00e9. D&rsquo;autres recherches dans ce domaine seront n\u00e9cessaires, mais deux choses sont claires : l&rsquo;amplitude, ou le volume, de nos rythmes circadiens semble \u00eatre extr\u00eamement importante pour la sant\u00e9, et cette amplitude est au moins partiellement contr\u00f4l\u00e9e par la prot\u00e9ine REV-ERB\u03b1.<\/p>\n<p>Le rythme circadien fait partie int\u00e9grante de la sant\u00e9 de tous les organismes, il est donc logique qu&rsquo;il soit \u00e9troitement contr\u00f4l\u00e9 \u00e0 tous points de vue.
