S\u00e9rotonine. <a href=\"https:\/\/www.chronobiology.com\/sleep-2\/melatonin\/\" target=\"_blank\" rel=\"noopener\">M\u00e9latonine<\/a>. La plupart des gens d&rsquo;aujourd&rsquo;hui connaissent ces neurotransmetteurs, qui jouent un r\u00f4le dans le fonctionnement de presque toutes les cellules de notre corps, en particulier dans nos cycles de sommeil. L&rsquo;inverse est \u00e9galement vrai : les personnes souffrant de troubles du rythme circadien semblent particuli\u00e8rement enclines \u00e0 adopter des comportements addictifs et \u00e0 abuser de substances.<\/p>\n<p>Afin d&rsquo;en savoir plus sur la nature de ce lien, les <a href=\"http:\/\/www.sciencedaily.com\/releases\/2005\/06\/050614001947.htm\" target=\"_blank\" rel=\"noopener\">chercheurs en chronobiologie ont \u00e9tudi\u00e9 deux groupes de souris<\/a>: Un groupe avec des mutations dans le g\u00e8ne de l&rsquo;horloge et un groupe avec un g\u00e8ne fonctionnant normalement. Les deux groupes de souris ont re\u00e7u de la coca\u00efne, une drogue qui cr\u00e9e une forte d\u00e9pendance, et ont ressenti les effets stimulants attendus. Cependant, le groupe de souris dont le g\u00e8ne de l&rsquo;horloge ne fonctionnait pas a commenc\u00e9 \u00e0 avoir un comportement \u00e9trange. Elles restaient dans la zone o\u00f9 elles avaient re\u00e7u de la coca\u00efne, m\u00eame lorsque des r\u00e9compenses leur \u00e9taient propos\u00e9es dans d&rsquo;autres zones. Elles montraient une pr\u00e9f\u00e9rence marqu\u00e9e pour la coca\u00efne et \u00e9taient pr\u00eates \u00e0 renoncer \u00e0 d&rsquo;autres activit\u00e9s gratifiantes pour avoir la chance d&rsquo;obtenir plus de drogue.<\/p>\n<p>La seule diff\u00e9rence significative entre les deux groupes de souris \u00e9tait le g\u00e8ne Clock, ce qui sugg\u00e8re que ce g\u00e8ne affecte le comportement de recherche du plaisir.<\/p>\n<h2>Dopamine, rythmes circadiens et d\u00e9pendance<\/h2>\n<p>L&rsquo;\u00e9tude du cerveau des souris a r\u00e9v\u00e9l\u00e9 plusieurs diff\u00e9rences essentielles entre celles qui poss\u00e9daient un g\u00e8ne Clock fonctionnel et celles qui n&rsquo;en poss\u00e9daient pas. Les neurones producteurs de dopamine des souris d\u00e9ficientes en horloge s&rsquo;activaient plus fr\u00e9quemment, ce qui se traduisait par un taux de dopamine plus \u00e9lev\u00e9. La dopamine est un neurotransmetteur cl\u00e9 impliqu\u00e9 dans les comportements de risque et de r\u00e9compense ainsi que dans le plaisir. Ces souris pr\u00e9sentaient un taux de dopamine plus \u00e9lev\u00e9 lorsqu&rsquo;elles \u00e9taient sobres, mais aussi une augmentation consid\u00e9rable lorsqu&rsquo;on leur donnait de la coca\u00efne. Il en r\u00e9sulte que les souris pr\u00e9sentant des troubles g\u00e9n\u00e9tiques du rythme circadien \u00e9prouvent plus de plaisir \u00e0 consommer de la coca\u00efne.<\/p>\n<h2>Abus d&rsquo;alcool et troubles du sommeil et de l&rsquo;\u00e9veil<\/h2>\n<p>Le g\u00e8ne de l&rsquo;horloge n&rsquo;est pas le seul g\u00e8ne du rythme circadien \u00e0 avoir \u00e9t\u00e9 mis en \u0153uvre dans l&rsquo;addiction. Une autre \u00e9tude a port\u00e9 sur le <a href=\"http:\/\/dartmed.dartmouth.edu\/fall13\/html\/advances_circadian\/\" target=\"_blank\" rel=\"noopener\">rythme circadien et l&rsquo;abus d&rsquo;alcool chez la souris<\/a>. Les souris qui pr\u00e9sentaient une mutation du g\u00e8ne Per, un autre g\u00e8ne li\u00e9 aux cycles veille-sommeil, \u00e9taient susceptibles de boire plus d&rsquo;alcool et de le m\u00e9taboliser plus lentement. Les souris porteuses de cette mutation pr\u00e9sentaient ce que l&rsquo;on appellerait un comportement de consommation excessive d&rsquo;alcool chez l&rsquo;homme, alors que les souris sans mutation \u00e9taient plus enclines \u00e0 consommer de l&rsquo;alcool avec mod\u00e9ration.<\/p>\n<h2>Qu&rsquo;est-ce que cela signifie pour les personnes souffrant de troubles circadiens ?<\/h2>\n<p>Les humains ne sont pas des souris, mais nous avons des <a href=\"http:\/\/www.ncbi.nlm.nih.gov\/pubmed\/24731209\" target=\"_blank\" rel=\"noopener\">g\u00e8nes et un comportement similaires dans le domaine du rythme circadien<\/a>. M\u00eame avant ces \u00e9tudes, de nombreux \u00e9l\u00e9ments sugg\u00e9raient que le cycle veille-sommeil \u00e9tait li\u00e9 \u00e0 la d\u00e9pendance. Par exemple, les <a href=\"http:\/\/www.psychologyinaction.org\/2009\/01\/04\/its-drug-time-circadian-rhythms-and-addiction\/\" target=\"_blank\" rel=\"noopener\">toxicomanes ont tendance \u00e0 avoir des troubles du sommeil<\/a> longtemps apr\u00e8s le sevrage d&rsquo;une drogue ; l&rsquo;intensit\u00e9 de ces troubles peut m\u00eame pr\u00e9dire si la personne va rechuter.
